Eldest son of Maj. Arthur Chamberlin and Rosanna Slaught, of Amwell, Washington Co., PA. In 1832, he married Clarissa Griffin. Clarissa and Robert had ten childdren: James; Geo. W.; Morgan; Nancy M.; Simon; John; Clarissa Jane (Jane); Sidney R. (female); Martha A.; and Robert. Clarissa (Griffin) Chamberlin died in 1859, and in 1861 Robert took Loraine Susannah (Horn) Riggle as his second wife. Robert and Loraine had no additional children.
